Carl Hiaasen, early John Irving…I started listing comedic writers, then read your “buddy adventure fantasy” requirement.
Hitchhikers Guide to the Galaxy
Fuck it, I am going back to authors who should satisfy your comic needs.
Tom Robbins, Kurt Vonnegut.

Buddy adventure fantasy comedy is a tall order, but a few that might work would be Kings of the Wyld by Nicholas Eames. It’s about an adventuring mercenary band gettnig back together for one last job. Plays into a bunch of fantasy tropes in a fun way. Another would be anything by Terry Pratchett but specifically Mort or Guards! Guards!

If it’s okay for the buddy to be Coyote the Trickster God, try Coyote Blue by Christoper Moore
Definitely adventure, fantasy, comedy. MC is on the hike alone, but periodically has some (almost) buddies along – including a potty-mouthed talking crab named Crab. How can you pass that up??
Ertata on
MythAdventures series by Robert Asprin
Neona65 on
Medium Dead by Christopher Dolley
It’s comedy fantasy adventure with a duo.
IntenseGeekitude on
Bridge of Birds by Barry Hughart
Also a second vote for Robert Asprin’s Myth Adventures series.

These are stand-alone, guy-centric ones:
Lamb by Christopher Moore
Gil’s All Fright Diner by A. Lee Martinez

Whatever you do, don’t read The Road.
Not fantasy but check Big Maria by Johnny Shaw.
[The Hike by Drew Magary.](https://www.goodreads.com/book/show/27833803-the-hike?ac=1&from_search=true&qid=n9Ts4A53Fe&rank=2)
